In order to ensure the correct collection, transportation and storage of specimens, Laboratory Science has compiled operating procedures for the collection, transportation, and storage of clinical laboratory specimens, requiring medical staff to strictly follow the operating procedures for collection, transportation and preservation of specimens.
1. Sample collection:
All specimen collection; transportation and processing should be carried out under the principle of aseptic operation; prevention of contamination.
All collected specimens should be placed in leak-proof; transported in sealed sterile containers;
Collect enough specimens.
Each specimen should be marked with the name of the patient; the number to be examined; the source of the specimen; the specific location; the date; the time and relevant clinical information.
Whole blood
When whole blood is used as the specimen to be tested; attention must be paid to the choice of anticoagulant; EDTA-Na2 or sodium citrate is generally used; heparin cannot be used
If the whole blood sample is used for DNA extraction and testing; it can be stored at 4℃ for a short time, if it is used for RNA testing; it should be taken after the blood is taken; RNA should be extracted as soon as possible.
Serum/plasma
DNA determination; can follow the general procedures of serum specimen processing; has little effect on the determination of RNA determination; the way the specimen is obtained and stored has a decisive influence on the determination result. It is best to use EDTA anticoagulation (heparin is strictly prohibited) whole blood specimens; plasma is separated within 6 hours after anticoagulation; if serum specimens are used, serum must be separated as soon as possible (within 2 hours); short-term specimens (1-2 weeks) It can be stored at -20°C, and it should be stored at -70°C for a longer period of time.

2. Delivery of samples
Once the sample is collected, it should be sent to the testing laboratory as soon as possible
If appropriate stabilizer is added to the sample; such as EDTA anticoagulant used for DNA determination, etc.; it can be shipped or mailed at room temperature
Recommendation: Express delivery of whole blood samples or DNA samples in ice packs and foam boxes. Samples that need to extract RNA need to be pre-processed and shipped with 1ml Trizol at low temperature.
Second, the preservation of samples
Routine peripheral blood samples; routine blood samples after collection; within 24 hours at room temperature; can be stored for 1 week at 4°C; for hemoglobin electrophoresis at room temperature for 1 week; electrophoresis can be stored at 4°C for 15 days; DNA extraction samples can be stored at room temperature for 24 hours; 1 sample at 4°C Weeks; -20°C for 3 months; -70°C for half a year to 3 years; RNA extraction samples at room temperature for 6 hours; 4°C for 12 hours; -70°C for 3 months; Amniotic fluid can be stored at 12 hours by centrifugation as above.
